چکیده

Hortic. bras., v. 22, n. 2, abr.-jun. 2004 C potato (Solanum tuberosum L.) is one of the most important vegetable crop of Brazil and the world’s fourth most important crop (Solomon-Blackbourn & Barker, 2001). In 2002, Brazil produced 3,126,411 t of potato with an average yield of 19,4 kg/ ha (IBGE, 2002). Potato is a tetraploid species and the use of its botanical seeds in commercial cultivation is precluded by low germinability and large variability in the segregant generations (Gardner & Snustad, 1986). Synthetic seed is one of the alternatives to solve this problem. The original definition of synthetic seeds, “a single encapsulated somatic embryo”, was given by Murashige (1978). In this concept, the synthetic BORDALLO, P.N.; SILVA, D.H.; MARIA, J.; CRUZ, C.D.; FONTES, E.P. Somaclonal variation on in vitro callus culture potato cultivars. In Vitro Techniques and Mutagenesis for the Genetic Improvement of Potato Cvs. Desiree and Diamant

The aim of the present study was to get somaclonal variants and induced mutants of potato for desirable characters with special emphasis on yield and yield components in two cultivars of potato viz., Desiree and Diamant. Inter-nodal explants from both the cultivars were incubated for 1420wks in callus inducing medium comprising MS salts supplemented with NAA (1.0mg/l) and BAP (0.5mg/l) for obta...

Detection of Somaclonal Variation in Potato Callus Induced by Uv-c Radiation Using Rapd-pcr

In this study, calli were obtained from in vitro grown potato stem or leaf segments cv. Cosima on MS medium containing 2,4-D, NAA, kinetin and yeast extract. After three subcultures, well grown calli were exposed to UV-C radiation with approximately 6.24 μmol photon/m2/s for 30 min three times at an interval of 2 weeks. Somaclonal variation induced by UV-C radiation was revealed by 5 out of 28 ...

Agrobacterium tumefaciens-mediated transformation of potato and analysis of genomic instability by RAPD

In order to enhance the system of potato transformation and further regeneration, four potato cultivars were transformed using the Agrobacterium tumefaciens harboring â-glucuronidase (GUS) gene. The results revealed that the highest percentages of shoot formation obtained were for Spunta cv. Stem explants resulted in more callus and regeneration after transformation and selection than leaf expla...

Regeneration of multiple shoots from transgenic potato events facilitates the recovery of phenotypically normal lines: assessing a cry9Aa2 gene conferring insect resistance

BACKGROUND The recovery of high performing transgenic lines in clonal crops is limited by the occurrence of somaclonal variation during the tissue culture phase of transformation. This is usually circumvented by developing large populations of transgenic lines, each derived from the first shoot to regenerate from each transformation event. This study investigates a new strategy of assessing mul...
